摘要

Indonesia as a megabiodiversity country have to explore biodiversity from the ecosystem level to genetic variations to avoid loss of them. Parijoto is ones of Indonesian plant used for ornamental plants and medicine. Parijoto included in genus of Medinilla. Ten species has been identified scattered on the mountains of Java. There are two cultivated species (Medinilla javanensis and M. verrucosa) in Muria mountainous, Central Java.Both of them have similar morphological character, therefore they often make confuses. On the other side, the genetic information of them is needed. Hence, on the level of gene is necessary to identify M. javanensis and M. verrucosa by molecular markers. The purposes of identification using molecular markers are to distinguish both of two spesies and get their genetic information. Identification on a level genes carried by variation sequence of DNA (polymorphism) between M. javanensis and M. verrucosa. Method of Restriction Fragment Length Polymorphism (RFLP) and Random Amplified Polymorphism DNA (RAPD) are usefull to identify genetic variation between organisms. The use of them should show polymorphism after observed using electrophoresis method. The appearance of polymorphism shows variations on the genetic level to strength the identification through morphological character. Result of the research will be applied as a learning material of high school students to explain genetic diversity. Students expected to understand genetic variations not only as an image on book, but in contextual way.
